Patrick Brown's Ultimate Soil Extract (SeX medium)
note: this
medium takes at least 10 days to make properly, always plan ahead.
Good Quality Garden
Soil 50g
NH4MgPO4-6H2O
200 mg
CaCO3
200 mg
Ground
Barley
200 mg
Dried Split
Peas
10 pieces
Heat until just before boiling (c.a. 85 °C), let sit uncovered at
room temperature overnight and repeat heating on the second day.
Let the mixture stand for 4-7 days uncovered at room temperature.
It is important to let the natural bacterial flora thrive for some time
to break down the nutrients in the soil, barley, and peas. Once
the mixture has the smell of good compost (slightly decayed but not
rotten). Carefully decant off the supernatant through cheesecloth
without disturbing the settled soil etc. into 500mL screw-top
bottles. Autoclave the resulting soil extract for at least 30
min. Let it sit for 7-10 days to allow for all the dead bacterial
cells to settle to the bottom of the bottle. Finally add 1 uL of
ES vitamin mix and dispense into sterile culture flasks, tubes,
bottles, etc.
